dc.description.abstract | The current work is divided into three main axes/topics: I) the calculation of residual stresses generated during tumor growth, II) the modeling of the effect of CAR-T cell therapy and anti-angiogenic treatments on tumor progression and III) the mathematical analysis of the role of the gut microbiome in cancer immunotherapy.
Firstly, an experimental and computational approach was developed to quantify growth-induced residual stresses in solid tumors, focusing on breast, pancreatic, and fibrosarcoma tumor models. The study reveals a non-uniform distribution of residual stresses within the tumor microenvironment, where the average magnitudes of residual stresses are increased after considering the swelling of the 2nd cut. Spatial analysis demonstrates strong associations between hyaluronan and collagen content and the spatial profile of stress, particularly in the more fibrotic tumor types. Furthermore, non-linear correlations between tumor volume and elastic modulus highlight the complex mechanical behavior of tumors. These findings provide valuable insights into the mechanical characteristics of solid tumors, enhancing our understanding of tumor progression and treatment response. This work was published in a first-author publication in Biomechanics and Modeling in Mechanobiology, 2023 Oct;22(5):1625-1643. doi: 10.1007/s10237-023-01716-3. Also, the findings of this research have been incorporated in an article accepted for publication in the AACR journal Clinical Cancer Research.
Secondly, a mechanistic mathematical model was developed to explore the complex interplay between tumor growth, angiogenesis, anti-angiogenic drugs, and chimeric antigen receptor T-cell (CAR-T) therapy. By integrating experimental data, the study identifies three primary mechanisms by which anti-angiogenic drugs influence tumor progression and treatment outcomes: reduction in tumor growth rate, normalization of blood vessel structure and function, and enhancement of immune cell infiltration and activation. The study underscores the significance of the normalization window for optimizing treatment benefits and highlights the potential of combining anti-angiogenic drugs with CAR-T cell therapy to improve patient outcomes. Furthermore, the study emphasizes the importance of carefully considering CAR-T cell dosing and dosing intervals to minimize adverse effects while maximizing therapeutic efficacy. These findings lay the groundwork for further research into optimizing CAR-T cell therapy and combination treatments for solid tumors. This work is in revision in the journal npj Biological Physics and Mechanics, a revised manuscript has been submitted and a final decision is pending.
Thirdly, a mathematical model based on mechanistic modeling of antitumor immune response, incorporating preclinical and clinical microbiome data was employed to investigate the role of the gut microbiome in modulating response to cancer immunotherapy. The study identifies activation and killing rate constants of immune cells as key determinants influenced by microbiome relative abundance. Association analysis reveals significant effects of specific bacterial families on the killing rate of immune cells, emphasizing the microbiome's impact on immunotherapy efficacy. These findings provide valuable insights into the underlying mechanisms by which the microbiome influences cancer immunotherapy response, offering a framework for optimizing treatment strategies through microbiome modulation. Further research guided by these insights has the potential to improve patient outcomes in cancer immunotherapy. | en |
dc.description.abstract | Η παρούσα εργασία διαιρείται σε τρεις κύριους άξονες: Ι) τον υπολογισμό των παραμένουσων τάσεων που προκαλούνται κατά τη διάρκεια της ανάπτυξης καρκινικών όγκων, ΙΙ) την μοντελοποίηση της αποτελεσματικότητας της θεραπείας με CAR-T κύτταρα σε συνδυασμό με αντιαγγειογενής θεραπεία και ΙΙΙ) την μελέτη του ρόλου του μικροβιώματος στην ανοσοθεραπεία του καρκίνου.
Πρώτα, αναπτύχθηκε μια πειραματική και υπολογιστική προσέγγιση για τον ποσοτικό προσδιορισμό των παραμένουσων τάσεων που προκαλούνται από την ανάπτυξη του συμπαγής καρκίνου, σε μοντέλα όγκων του μαστού, παγκρέατος και ινοσαρκώματος. Η μελέτη αποκαλύπτει μια μη ομοιόμορφη κατανομή των παραμένουσων τάσεων εντός του μικροπεριβάλλοντος του όγκου, με τις μέσες τιμές των παραμένουσων τάσεων να αυξάνονται όταν συνυπολογιστεί και η διόγκωση μετά από την δεύτερη τομή. Η χωρική ανάλυση δείχνει ισχυρές συσχετίσεις μεταξύ του υαλουρονικού οξέος, κολλαγόνου και του προφίλ των τάσεων, ιδιαίτερα στους πιο ινοτικούς τύπους όγκων. Επιπλέον, η μη γραμμική συσχέτιση μεταξύ του όγκου του καρκίνου και του μέτρου ελαστικότητας υπογραμμίζει την πολύπλοκη μηχανική συμπεριφορά των όγκων. Τα ευρήματα αυτά παρέχουν πολύτιμες πληροφορίες σχετικά με τα μηχανικά χαρακτηριστικά των συμπαγών όγκων, ενισχύοντας την κατανόησή μας σχετικά με την εξέλιξη του όγκου και την ανταπόκριση στη θεραπεία.
Δεύτερον, αναπτύχθηκε ένα μηχανιστικό μαθηματικό μοντέλο για την διερεύνηση των πολύπλοκών αλληλεπιδράσεων της ανάπτυξης του καρκίνου, της αγγειογένεσης, των αντιαγγειογενών φαρμάκων και της θεραπείας με T λεμφοκύτταρα που φέρουν χιμαιρικούς αντιγονικούς υποδοχείς (CAR-T κύτταρα). Με την συμβολή πειραματικών δεδομένων, η μελέτη αναδεικνύει τρεις κύριους μηχανισμούς με τους οποίους τα αντιαγγειογενοί φάρμακα επηρεάζουν την εξέλιξη του καρκίνου και την αποτελεσματικότητα της θεραπείας: μείωση του ρυθμού ανάπτυξης του καρκίνου, επιδιόρθωσης της δομής και λειτουργίας των αιμοφόρων αγγείων, και ενίσχυση της εισροής και ενεργοποίησης των ανοσοποιητικών κυττάρων. Η μελέτη υπογραμμίζει τη σημασία του περιθωρίου βέλτιστης επιδιόρθωσης από την αντιαγγειογενή θεραπεία και το συνδυασμό με θεραπεία CAR-T για επίτευξη βέλτιστου αποτελέσματος. Επιπλέον, η μελέτη τονίζει τη σημασία της δοσολογίας των CAR-T κυττάρων και των μεσοδιαστημάτων δοσοληψίας για την ελαχιστοποίηση των ανεπιθύμητων παρενεργειών και ταυτόχρονη μεγιστοποίηση της αποτελεσματικότητας της θεραπείας. Τα ευρήματα αποτελούν τα θεμέλια για περαιτέρω έρευνα στη βελτιστοποίηση της θεραπείας με κύτταρα CAR-T και συνδυασμό άλλων θεραπειών.
Τρίτον αναπτύχθηκε ένα μαθηματικό μοντέλο βασισμένο στη μηχανιστική μοντελοποίηση της αντίδρασης των κυττάρων του ανοσοποιητικού κατά του καρκίνου, με χρήση προκλινικών και κλινικών δεδομένων εντερικού μικροβιωμάτος για τη διερεύνηση του ρόλου του στην ανταπόκριση στην ανοσοθεραπεία του καρκίνου. Η μελέτη εντόπισε τις παραμέτρους ενεργοποίησης και κυτταροτοξικότητας των κυττάρων του ανοσοποιητικού ως κύριους παράγοντες που επηρεάζονται από τον σχετικό πληθυσμό των μικροβίων. Η ανάλυση συσχέτισης αποκαλύπτει σημαντικές επιδράσεις συγκεκριμένων οικογενειών βακτηριών στη παράμετρο κυτταροτοξικότητας των κυττάρων του ανοσοποιητικού, τονίζοντας την επίδραση του μικροβιώματος στην αποτελεσματικότητα της ανοσοθεραπείας. Αυτά τα ευρήματα παρέχουν πολύτιμη συνεισφορά στους υποκείμενους μηχανισμούς με τους οποίους το μικροβίωμα επηρεάζει την ανταπόκριση στην ανοσοθεραπεία του καρκίνου, προσφέροντας ένα πλαίσιο για τη βελτιστοποίηση της θεραπείας του καρκίνου μέσω του μικροβιώματος. Τα συμπεράσματα αυτά μπορούν να καθοδηγήσουν περαιτέρω πειραματική έρευνα για τη βελτιστοποίηση της ανοσοθεραπείας του καρκίνου. | el |
