dc.description.abstract | Kύριος στόχος της παρούσας διατριβής ήταν η ανάπτυξη και η επικύρωση μιας UPLC-MS/MS μεθόδου για την ποσοτικοποίηση κανναβινοειδών ενώσεων στο κερί των αυτιών από χρήστες κάνναβης. Μετά την ανάλυση των δειγμάτων παρατηρήθηκε σημαντική διαφορά στην ποσότητα του παραγόμενου κεριού και τις συγκεντρώσεις των κανναβινοειδών που εντοπίζονταν στο κερί των χρηστών. Ακόμη, η εν λόγω διατριβή, είχε ως στόχο την ανάπτυξη μιας πρόσθετης μεθόδου LC-MS, για την ποσοτικοποίηση κανναβινοειδών ενώσεων σε τρόφιμα κάνναβης που διατίθενται προς πώληση στην Κυπριακή αγορά. Διάφορες διαδικασίες εκχύλισης δοκιμάστηκαν, όπως η SPE, η εκχύλιση Soxhlet και η UAE, με την UAE να αποδεικνύεται πιο αποτελεσματική για στερεά τρόφιμα κάνναβης, ενώ η εκχύλιση SPE, είχε παρόμοια αποτελέσματα με την UAE, για υγρά δείγματα τροφίμων. Κατά την ανάλυση, παρατηρήθηκε ότι το τσάι κάνναβης εμφάνισε τη μεγαλύτερη συγκέντρωση κανναβινοειδών, οδηγώντας στην επιλογή του για περαιτέρω πειραματισμό. Έτσι λοιπόν, αναπτύχθηκε και βελτιστοποιήθηκε μια τεχνική εκχύλισης MAE για την ανάκτηση πέντε κανναβινοειδών ενώσεων από τσάι κάνναβης χρησιμοποιώντας την μεθοδολογία επιφάνειας απόκρισης (RSM). Οι παράμετροι της θερμοκρασίας, του χρόνου εκχύλισης και της αναλογίας διαλύτη προς την ποσότητα τσαγιού εξετάστηκαν και τρισδιάστατα διαγράμματα που καταδείκνυαν τις βέλτιστες συνθήκες εκχύλισης δημιουργήθηκαν. Επιπλέον, μια ακόμη βελτιστοποίηση με RSM πραγματοποιήθηκε χρησιμοποιώντας τη διαδικασία UAE με ιοντικά υγρά (ILs). Αυτή τη φορά, οι βελτιστοποιημένες συνθήκες του χρόνου εκχύλισης, της συγκέντρωση διαλύτη, και της αναλογίας τσαγιού προς τον όγκο του διαλύτη εξετάστηκαν. Όλα τα πιο πάνω πειράματα κατέδειξαν το CBD ως το κυρίαρχο κανναβινοειδές στα τρόφιμα κάνναβης, με όλα τα ελεγμένα τσάγια να περιέχουν σημαντικά επίπεδα Δ9-ΤΗC σε μεταβαλλόμενες ποσότητες. | el |
dc.description.abstract | The primary objective of the current thesis was to develop and validate a UPLC-MS/MS method for quantifying cannabinoids in cerumen from cannabis users. The analysis of samples from cannabis users revealed notable inter-individual variability in cannabinoid concentrations and cerumen production. Moreover, the research aimed to develop an additional LC-MS method to analyze cannabinoids in cannabis-based edibles that are available in Cyprus market. Various extraction procedures, including SPE, Soxhlet extraction, and UAE, were evaluated, with UAE proving to be more effective for solid food products while the SPE procedure proved to be comparable to UAE for liquid samples. Upon analysis, it was observed that hemp tea exhibited the highest concentration of cannabinoids among the examined cannabis-infused edibles, leading to its selection for further experimentation. Subsequently, MAE technique was examined and optimized for extracting five cannabinoids from hemp tea using response surface methodology (RSM). The parameters of temperature, extraction time, and solvent-to-solid ratio were investigated and the obtained results were expressed in the form of 3D plots. Furthermore, another RSM optimization was conducted by utilizing the UAE procedure with ionic liquids (ILs). This time, the optimized conditions of extraction time, solvent concentration, and solid to solvent ratio were tested. The analysis unveiled CBD as the most prevalent cannabinoid in all cannabis products, with all tested hemp teas containing significant levels of Δ9-ΤΗC in varying quantities. | en |
